A Comparative Analysis of Somatic and Excretory-Secretory Proteins of Haemonchus contortus and Ostertagia ostertagi Infecting Sheep of Kashmir Valley

چکیده
Helminth parasitism of food and dairy animals has been responsible for significant economic losses throughout the world. Although a number of helminth parasites affect ruminants, but Haemonchus contortus and Ostertagia ostertagi are of paramount importance as they affect their hosts in a number of ways and often result in their death. The aim of the present trial was to do a comparative analysis of somatic as well as the excretory-secretory proteins of both the parasites. Many significant protein bands were found in case of somatic as well as the excretory-secretory proteins of both the parasites which will play an important role in controlling these two parasites. E/S antigens were found to be more in number as compared to somatic antigens. This means E/S antigens are more antigenic as compared to the somatic antigens. Further 2D gel electrophoresis and mass spectrometric analysis of these diagnostic E/S antigens are needed which will identify the particular proteins and thereby will play an important role in devising an effective vaccine against these parasites.

Immunoprotection in sheep against Haemonchus contortus using its thiol-purified excretory/secretory proteins
Excretory/Secretory antigen was prepared by culturing live adult worms of Haemonchus contortus in RPMI 1640 medium at a concentration of 50 worms per mL in a culture-flask at 37 ˚C for 24 hr and the culture supernatant was used as antigen. The E/S antigen was purified by thiol-sepharose affinity chromatography. On western blot analysis, it was demonstrated that thiol-purified antigen showed a s...

Indirect plate enzyme-linked immunosorbent assay was standardized and evaluated for its effectivenessin immunodiagnosis of haemonchosis in experimental and clinical cases in sheep by using somatic wholeadult antigen of H. contortus. Plate ELISA was standardized using 5 μg/well antigen concentration with1:100 and 1:1000 of sera and conjugate dilution. Indirect plate ELISA was able to demonstrate...

Exsheathing fluid (EF) and excretory-secretory products (ES) of infective third-stage cultured larvae ofOstertagia circumcincta were analysed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDSPAGE). Five and seven predominant proteins were found in the EF and ES products, respectively. Immunoblotting by sheep pre-infection serum did not react with any of the EF and ES proteins, b...

Two doses of infective larvae of 20 000 Trichostrongylus axei, dosed to Merino lambs at an interval of 14 days and subsequently challenged with Ostertagia circumcincta, caused a significant reduction (P less than 0.01) in the establishment of 5th and adult stages of the latter. T. axei was unable to protect Merino sheep against homologous challenge nor was Haemonchus contortus a successful vacc...
